The first elutingfactions (tR1) were concentrated to dryness, and washed withsaturated aqueous NaHCO3, extracted with EtOAc. The organic layer was separated, washed with water and saturatedaqueous NaCl, dried over anhydrous MgSO4 and concentratedin vacuo. The residue (42.7 mg) was crystallized from hexane-ethyl acetate to give 17 (34.4 mg, 0.072 mmol, 7.2%) asa white solid. 1H-NMR (300 MHz, DMSO-d6) delta: 1.04 (3H, s),1.37 (3H, s), 2.44 (3H, s), 4.87 (1H, d, J=8.3 Hz), 5.39 (1H, s),7.23-7.34 (2H, m), 7.48-7.58 (2H, m), 7.93 (1H, d, J=7.5 Hz),8.55 (1H, s), 8.88 (1H, d, J=0.8 Hz), 9.07 (1H, d, J=8.3 Hz),9.50 (1H, d, J=7.5 Hz). MS (ESI/APCI) m/z 476.2 [M+H]+.HPLC purity: 99.4%. mp 260C.

To a flask containing 1-methyl-1H-1,2,3-triazole (200 mg, 2.41 mmol) was added THF (15 mL) and the solution was cooled to -45 C. using a CH3CN-CO2 bath. n-BuLi (2.5 M in hexanes, 1.0 mL, 2.5 mmol) was added dropwise which afforded an opaque white suspension. The mixture was stirred at -45 C. for 25 minutes, then a solution of (4-chloro-3-cyclopentyl-2-methoxyquinolin-6-yl)(1,2-dimethyl-1H-imidazol-5-yl)methanone (475 mg, 1.24 mmol, Intermediate 68: step c) in THF (3 mL) was introduced. After 10 minutes, the -45 C. bath was replaced with an ice-water bath. After 30 minutes, the reaction mixture was quenched with aqueous NH4Cl solution and extracted with EtOAc:THF (2:1, 3*40 mL). The combined organics were washed with brine, dried over MgSO4, filtered and concentrated. Chromatography on silica gel (3% MeOH-DCM increasing to 10% MeOH) provided the title compound as a light tan solid. 1H NMR (500 MHz, CDCl3) delta ppm 8.24 (d, J=1.9 Hz, 1H), 7.69 (d, J=8.7 Hz, 1H), 7.35 (dd, J=8.7, 2.0 Hz, 2H), 7.07 (s, 1H), 6.01 (s, 1H), 4.07 (s, 3H), 3.90-3.80 (m, 4H containing a 3H singlet at 3.86), 3.32 (s, 3H), 2.06-1.94 (m, 3H), 1.94-1.84 (m, 6H), 1.75-1.69 (m, 2H). MS (ESI): mass calcd. for C24H27ClN6O2, 466.2. m/z found 467.1 [M+H]+. The racemate was separated by chiral SFC (Stationary phase: Chiralpak AD-H, 5 mum 250¡Á20 mm, Mobile phase: 70% CO2: 30% EtOH (0.3% iPrNH2)) to give Example 135b as the first compound that eluted from the chiral column and Example 135c as the second compound that eluted from the chiral column.

The reaction progress was monitored by HPLC and the reaction was judged complete after two hours. The reaction was quenched with H20 (250 mL) and the pH- value was adjusted to pH = 10.0 by the addition of NaOH (2.0 M).The organic layer was separated and washed with H20 (250 mL) at a pH = 2.0 adjusted with HC1 (2.0 M). The organic phase was concentrated under reduced pressure. The residue was taken up in toluene (100 mL) and concentrated under reduced pressure to give 49.6 g residue. The residue was taken up in cyclohexane (~ 250 mL) and stirred for two hours at room temperature. The solid was filtered, washed twice with cyclohexane (2 x 20 mL) and dried (40 C, 1H NMR (300 MHz, DMSO-D6): delta = 6.77 - 8.07 (series of overlapping m, 6H), 3.88 - 4.37 (series of overlapping m, 1H), 3.42 - 3.66 (three s, 3H), 2.56 - 3.35 (series of overlapping m, 6H), 2.39 (three s, 3H), 0.82 - 1.40 (series of overlapping m, 12H). The combined organic layers were dried over magnesium sulfate, filtered, and concentrated onto Celite. Purification by flash column chromatography using 0-40% EtOAc/hexanes as eluent provided the title compound as an off-white solid (9.65 g, 52%): mp 109-112 C.; 1H NMR (400 MHz, CDCl3) delta 8.56 (s, 1H), 8.10-8.03 (m, 2H), 7.83-7.75 (m, 2H), 7.64-7.57 (m, 2H), 7.42-7.35 (m, 2H); ESIMS m/z 386 ([M+21-1 ]+).
