The crude material was purified by column chromatography using 15-25% EtOAc:hexane to afford 1-fluoro-2-methoxy-4-nitrobenzene (1.2 g, 35%) as an off-white solid. 1H-NMR (CDCl3, 400 MHz): delta 8.87 (s, 1H), 8.10 (d, 1H), 8.00-7.97 (m, 2H), 4.10 (s, 3H), 2.50 (s, 3H); LCMS: 234.9 (M+1); (column; Ascentis Express C-18 (50×3.0 mm, 2.7 mum); RT 2.02 min. 0.025% Aq TFA+5% ACN: ACN+5% 0.025% Aq TFA; 1.2 mL/min); TLC: 30% EtOAc:hexane (Rf: 0.2).

In a 50 mL round bottomed flask, 3,5-dibromo-1H-1,2,4-triazole (Int-1, 1.50 g, 6.61 mmol) was dissolved in DMF (15 mL) and the solution was cooled to 0-5 C (ice bath). Sodium hydride (55% dispersion in mineral oil, 317 mg, 7.93 mmol) was added in portions and the resulting mixture was stirred for 30 min at 0-5 C and for 15 min at room temperature. After that, 1- fluoro-2-iodoethane (1.15 g, 537 mu, 6.61 mmol) was added dropwise at room temperature. The resulting mixture was stirred for 18 h at room temperature. After that, it was concentrated in vacuo, the residue was purified by column chromatography (silica gel, 70 g, eluting with ethyl acetate / n-heptane, gradient 0: 100 to 40:60 v/v) to yield the title compound as yellow oil (1.51 g, 84%). 1H NMR (CDCl3, 300 MHz): delta 4.46 (td, J = 4.7, 24.2 Hz, 2 H), 4.80 (td, J = 4.7, 46.3 Hz, 2 H). MS (ES+) m/z 271.9, 273.9, 275.9 [M+H, 2 Br isotopes].

A mixture of 1-benzyl-1,2,4-triazole-3-carboxylic acid (16 mg, 0.08 mmol), (6S)-6-amino-2,4-dimethyl-7,8-dihydro-6H-pyrazolo[4,3-b]azepin-5-one (15 mg, 0.08 mmol), 1-(3-dimethylaminopropyl)-3-ethylcarbodiimidehydrochloride (15 mg, 0.08 mmol) and 1-hydroxybenzotriazole (10 mg, 0.08 mmol) in N,N-dimethylformamide (5 mL) was stirred at 30 C. for 2 h. The mixture was concentrated under reduce pressure. The residue was purified by RP-HPLC (acetonitrile 22-50%/0.05% hydrochloride acid in water) to afford 1-benzyl-N-[(6S)-2,4-dimethyl-5-oxo-7,8-dihydro-6H-pyrazolo[4,3-b]azepin-6-yl]-1,2,4-triazole-3-carboxamide (16.3 mg, 56%) as a white solid. 1H NMR (400 MHz, CD3OD) delta 8.81 (s, 1H), 7.84 (s, 1H), 7.38-7.33 (m, 5H), 5.51 (s, 2H), 4.73-4.68 (m, 1H), 3.93 (s, 3H), 3.31 (s, 3H), 3.00-2.90 (m, 2H), 2.50-2.45 (m, 1H), 2.27-2.22 (m, 1H). LCMS RT=1.088 min, m/z=380.2 [M+H]+. LCMS (5 to 95% acetonitrile in water+0.03% trifluoroacetic acid over 1.5 mins) retention time 1.088 min, ESI+ found [M+H]=380.2.
