This gave 38 mg (100.0% pure, 11.4% yield) of 6′-(3-chloro-1,2,4-triazol-1-yl)-[1(2H),3′-bipyridin]-2-one. Log P value (HCOOH)=1.23 LC-MS (ESI positive)=274.0 (M+) C12H8ClN5O (273.7 g/mol) 1H-NMR (400.0 MHz, DMSO-d6): delta=6.40; 6.55; 7.57; 7.79; 7.97; 8.22 (m, 6H, hetaryl-H); 8.67 (d, 1H, hetaryl-H); 9.51 (s, 1H, hetaryl-H) ppm.

To a stirred mixture of 5 (65 mg, 0.173 mmol), 1 – 1 ,2,3-triazoleacetic acid (6, 22 mg, 0.173 mmol), and Hunig’s base (0.091 mL, 0.522 mmol) in DMF (2 ml) was added HATU (66 mg, 0.174 mmol) and the reaction mixture was stirred for 30 min. MS (EI) for C24H27N, ,0, found 486 (MH+).

The combined organic phases were dried over anhydrous MgSO4, filtered, and concd in vacuo to provide 0.71 g of a light yellow oil (yield: 82%). The crude product was used without purification.

Isomer B 1H-NMR (400 MHz, CDCl3): 4.18 (s, 3H, Me) ppm.
