Herein, Fe3O4 nano-particles are anchored over nitrogen doped reduced graphene oxide Fe3O4@N-rGO) structure to accommodate electric and magnetic components, so that it could match both the conduction and polarization loss to augment the attenuation of electromagnetic radiation. The formation of condensed heteroaromatic structure and decoration of Fe3O4 particles on r-GO structure are confirmed by photoelectron spectroscopy (XPS). high-resolution transmission electron microscopy (HRTEM), X-ray diffraction (XRD). The Fe3O4@N-rGO hybrid is subsequently dispersed in epoxy matrix, which result in significantly higher electrical conductivity (1.31 s/m) and electromagnetic interference (EMI) shielding efficiency (- 26 dB at 1 mm thickness). It was assumed that presence of electric-magnetic integrated Fe3O4@N-rGO hybrid in epoxy matrix significantly improved the interfacial polarization and anisotropy energy, which was reflected in absorption loss value of -27 dB. The composites also exhibited enhanced mechanical and thermo-mechanical properties coupled with improved thermal stability.

Protein kinases orchestrate diverse cellular functions; however, their dysregulation is linked to metabolic dysfunctions, associated with many diseases, including cancer. Mitogen-Activated Protein (MAP) kinase is a notoriously oncogenic signaling pathway in human malignancies, where the extracellular signal-regulated kinases (ERK1/2) are focal serine/threonine kinases in the MAP kinase module with numerous cytosolic and nuclear mitogenic effector proteins. Subsequently, hampering the ERK kinase activity by small molecule inhibitors is a robust strategy to control the malignancies with aberrant MAP kinase signaling cascades. Consequently, new heterocyclic compounds, containing a sulfonamide moiety, were rationally designed, aided by the molecular docking of the starting reactant 1-(4-((4-methylpiperidin-1-yl)sulfonyl)phenyl)ethan-1-one (3) at the ATP binding pocket of the ERK kinase domain, which was relying on the molecular extension tactic. The identities of the synthesized compounds (4-33) were proven by their spectral data and elemental analysis. The target compounds exhibited pronounced anti-proliferative activities against the MCF-7, HepG-2, and HCT-116 cancerous cell lines with potencies reaching a 2.96 mu M for the most active compound (22). Moreover, compounds 5, 9, 10b, 22, and 28 displayed a significant G2/M phase arrest and induction of the apoptosis, which was confirmed by the cell cycle analysis and the flow cytometry. Thus, the molecular extension of a small fragment bounded at the ERK kinase domain is a valid tactic for the rational synthesis of the ERK inhibitors to control various human malignancies.

For the first time, the coordination-based energy sacrificial bonds have been constructed in the interface between lignin and polyolefin elastomer for preparing a new class of high performance thermoplastic elastomers (TPEs) with biomass lignin as the hard plastic phase. The coordination bonds not only promoted the dispersion of lignin, but also improved the interfacial interactions between lignin and polyolefin elastomer matrix, and also facilitated the orientation of chain segments during stretching. The synergistic coordination effect of lignin promoted higher energy dissipation, leading to simultaneously enhanced strength and toughness of lignin-based TPEs even with the lignin loading as high as 30 wt %. The lignin-based TPEs also exhibited excellent shape memory performance. Our strategy offers a promising methodology for the facile production of high performance but cost-effective TPE materials using biorenewable resources as plastic phase.

Mitochondria are known as powerhouse of cells and play the role of a bridge in redox balance, cell apoptosis, and autophagy. ROS accumulation can cause mitochondria damage, while the injured mitochondria will further enhance ROS levels reciprocally. Herein, we synthesized a novel series of [1,2,4] triazolo[1,5-a]pyrimidine-based compounds 4a-4v and tested their anti-proliferation efficacy against gastric cancer cell line MGC-803. Among them, compounds 4o and 4p inhibited gastric cancer cells at micromolar level. Compound 4o caused G2/M arrest and induced mitochondria-dependent apoptosis in MGC-803 and SGC-7901. However, inhibiting apoptosis pathway cannot prevent the inhibitory activity of compound 4o against gastric cancer cell. To our surprising, ROS level was increased by compound 4o and elevation of ROS could be rescued by NAC. In accordance with that, NAC absolutely prevented the anti-proliferation efficacy of compound 4o. We further found that autophagy inhibitor CQ rather than 3-MA partially reversed inhibitory activity of compound 4o in MGC-803 cells. Taken together, compound 4o exhibited its anti-proliferative activity via increasing ROS level and inducing autophagy, thus leading to apoptosis of gastric cancer cells. In search of unprecedented tri and/or tetrapod pharmacophoric conjugates, a series of 32 new 4-ethyl-1H-benzo[b][1,4]diazepin-2(3H)-ones were synthesized and properly elucidated using MS, IR, NMR, and elemental analysis. In vitro investigation of 11 compounds of this series, using a panel of two human tumor cell lines namely; human breast adenocarcinoma (MCF-7), and human colorectal carcinoma (HCT-116), revealed promising cytotoxic activities. Among all synthesized compounds, analogue 9 displayed maximum cytotoxicity with IC50 values of 16.19 +/- 1.35 and 17.16 +/- 1.54 mu M against HCT-116 and MCF-7, respectively, compared to standard drug doxorubicin.

The Mbp1 protein functions as a DNA-binding protein in the MluI cell cycle box-binding complex and plays significant roles in yeast development. In this study, an ortholog of yeast Mbp1, BbMbp1, was characterized in a filamentous insect mycopathogen, Beauveria bassiana. BbMbp1 plays an important role in morphological changes under aerial and liquid environments. On the aerial surface, BbMbp1 was indispensable for the biogenesis of conidiophores and conidiation. Under submerged conditions, the increment BbMbp1 mutant displayed abnormal spore-producing structures, with a dramatic decrease in blastospore yield (similar to 95%). The virulence of the increment BbMbp1 mutant was notably weakened, which might be due to the defect in in vivo blastospore formation in the insect. Moreover, disruption of BbMbp1 resulted in a substantial reduction in hyphal growth on cadavers. Comparative transcriptomics revealed that BbMbp1 mediated different transcriptomes during the formation processes of conidia and blastospores. Yeast one-hybrid assays demonstrated that BbMbp1 was required for transcriptional control of a cell wall protein gene, BbCwp, and an integral membrane protein gene, BbImp that played significant roles in conidiation and blastospore formation respectively. Our results demonstrate that BbMbp1 contributes to the morphological transitions in the pathogenic and saprophytic growth of B. bassiana via different genetic pathways.

Developmentally-lead (Pb)-exposed rats showed an enhanced vulnerability to the stimulating and motivational effects of ethanol (EtOH). This is accompanied by differential activity of the brain EtOH-metabolizing enzymes catalase (CAT) and mitochondrial aldehyde dehydrogenase (ALDH2). Based on the theory that brain acetaldehyde accumulation is associated with the reinforcing properties of EtOH, this study sought to determine brain CAT and ALDH2 expression in limbic areas of control and Pb-exposed animals after voluntary EtOH intake. Thirty-five-day-old rats perinatally exposed to 220 ppm Pb were offered with water or increasing EtOH solutions (2-10% v/v) during 28 days until postnatal day (PND) 63. Once intake was stable, the animals were administered: 1) saline (SAL; test days 21-24 or 21-28, as corresponds), or 2) a CAT inhibitor: 3-amine 1, 2, 4-triazole (AT; 250 mg/kg intraperitoneally [i.p.], 5 h before the last eight EtOH intake sessions -test days 21-24 and 25-28), or 3) a CAT booster: 3-nitropropionic acid (3NPA; 20 mg/kg subcutaneously [s.c.], 45 min before the last four EtOH intake sessions -test days 25-28). Two additional groups were centrally-administered cyanamide (CY, an ALDH2 inhibitor, 0.3 mg i.c.v. immediately before the last four EtOH sessions, test days 25-28) or its corresponding vehicle (VEH). Lead exposure increased EtOH intake, an effect potentiated in both groups by 3NPA or CY pretreatments and reduced by AT, albeit selectivity in the Pb group. Catalase abundance in limbic areas parallels these observations in the Pb group, showing higher CAT expression in all areas after EtOH consumption respect to the controls, an effect prevented by AT administration. In contrast, ALDH2 expression was reduced in the Pb animals after EtOH intake, with CY potentiating this effect in all brain areas under study. Based on these results and on previous evidences, we suggest that Pb exposure promotes acetaldehyde accumulation in limbic regions, providing some insights into the mechanism of action that underlies the vulnerability to the excessive EtOH consumption reported in these animals.

Graphitic carbon nitride (g-C3N4) has been shown as a promising visible-light photosensitizer for photodynamic therapy (PDT) application. Nevertheless, its therapeutic efficiency is limited by the low efficiency of visible-light utilization. To overcome this issue, 3-amino-1,2,4-triazole-derived graphitic carbon nitride nanosheets (g-C3N5 NSs) are prepared for PDT application. The addition of nitrogen-rich triazole group into the g-C3N4 motif significantly makes the light absorption of g-C3N5 NSs red-shift with the band gap down to 1.95 eV, corresponding to a absorption edge at a wavelength of 636 nm. g-C3N5 NSs generate superoxide anion radicals (O-2(center dot-)) and singlet oxygen (O-1(2)) under the irradiation of a low-intensity white light emitting diode. Owing to the high efficiency of visible-light utilization, g-C3N5 NSs show about 9.5 fold photocatalytic activity of g-C3N4 NSs. In vitro anticancer studies based on the results of CCK-8 assay, Calcein-AM/PI cell-survival assay and photo-induced intracellular ROS level analysis in living HeLa cells demonstrate the potential of g-C3N5 NSs as a low-toxic and biocompatible high-efficient photosensitizer for PDT. A colorimetric and visual assay is described for the herbicide aminotriazole (ATZ). It is based on the etching of gold nanorods (AuNRs) by iodine which is formed on oxidation of iodide via H2O2. Longitudinal etching of the AuNRs occurs quickly and is accompanied by a color change from dark blue to red. In the absence of ATZ and the presence of active catalase (CAT), H2O2 is quickly decomposed into water, and the AuNRs will not be etched. In thepresence of ATZ, CAT is partially deactivated, and this affects the amount of available H2O2 and, consequently, of the iodine. Hence, the color is significantly changed. The color changes can be easily detected with bare eyes. The assay has a linear response in the 5 to 70 mu M concentration range, with a detection limit of 1.3 mu M and high selectivity for ATZ. It was applied to the determination of ATZ in water and food samples.
