Co-crystals are emerging as members of supramolecular family having particular practical and fundamental significance for chemists, crystallographers, pharmaceutical scientists, and theoreticians. The presented study is focused to synthesize anti-cancer co-crystals of commercially available nandrolone (1), a synthetic anabolic-androgenic steroidal drug. Co-crystallization is done using economical and green grinding and reflux methods to obtain nandrolone (Nan): salicylic acid (Sal) and nandrolone (Nan): 3-amino-1,2,4-triazole (Triz) co-crystals, in 2:1 and 1:1 stoichiometric ratios, respectively. The structural analysis and characterization were carried out using single-crystal X-ray diffraction, and vibrational spectroscopy. Nandrolone (1) crystallizes in monoclinic P2(1) space group, while the co-crystal-I (Nan:Sal) and co-crystal-II (Nan:Triz), co-crystallized in the orthorhombic P2(1)2(1)2(1) space group. The intermolecular hydrogen bonds O-H center dot center dot center dot O, C-H center dot center dot center dot O, N-H center dot center dot center dot O, O-H center dot center dot center dot N, and N-H center dot center dot center dot N between the active pharmaceutical ingredient (nandrolone) and co-formers (salicylic acid, and 3-amino-1,2,4-triazole) stabilize the structures of cocrystals. In vibrational spectroscopy of co-crystal-I (Nan:Sal), the blue shifts in stretching frequencies of hydroxyl group from 3417.9 cm(-1) to 3427.8 cm(-1) further supported the hydrogen bond interactions between API and co-former. Similarly, in co-crystal-II (Nan:Triz) the NH2 stretching frequency from 3331.4- 3413.4 cm(-1) to 3312.7 cm(-1), supported the interaction of NH2 with API via intermolecular interaction. Nandrolone (1) and both co-crystals were found to be non-cytotoxic against 3T3 normal fibroblast cell line. Nandrolone (IC50 = 1.0 0.1 mu M), co-crystal-I (Nan:Sal) (IC50 = 1.6 +/- 0.3 mu M) and-II (Nan:Triz) (IC50 = 1.8 +/- 0.1 mu M) showed anti-cancer potential against cervical cancer HeLa cell line. While doxorubicin (IC50 = 1.2 +/- 0.2 mu M) was used as standard tested compound. The formation of 4 entails selective and consecutive displacement of the 3-ethoxy and methoxy (ester) anions in I by 3-NH2 and 1-NH of 3-aminopyrazole. On the other hand, the formation of 7 implies selective displacement of 3-ethoxy in I by the ring-NH followed by cyclocondensation involving the keto group in I and 3-NH2 of aminotriazole. This latter selective displacement sequence is also followed by 3-amino-5-hydroxypyrazole in its reaction with I. The structures of the new compounds are supported by microanalytical and spectral data.

Availability and efficient utilization of host-derived nutrients by pathogens decide the fate of host-pathogen interaction. In Magnaporthe oryzae, N-acetylglucosamine (GlcNAc) catabolic pathway was found essential for successful host colonization and pathogenicity. GlcNAc catabolic enzymes hexokinase, GlcNAc-6-phosphate deacetylase (MoDac) and GlcN-6-phosphate deaminase (MoDeam) are encoded in a genomic cluster in M. oryzae and several phytopathogenic fungi. However, transcriptional regulation of GlcNAc catabolic pathway was not understood. We identified a conserved Ndt80/PhoG-like transcriptional regulator as a part of the GlcNAc catabolic gene cluster in M. oryzae and other fungi. We found that MoNdt80 is essential for GlcNAc utilization and pathogenicity of M. oryzae. Unlike WT, Delta MoNdt80 failed to induce transcription of GlcNAc catabolic pathway genes in response to GlcNAc. MoNdt80 could bind to a specific cis-acting consensus sequence GNCRCAAA[AT], present in the promoter of MoDac, MoDeam and beta-hexosaminidase (MoHex). Further, comparative RNA-sequencing analysis using WT and Delta MoNdt80 revealed a large set of GlcNAc responsive genes that are under the transcriptional control of MoNdt80. These genes encoded GlcNAc catabolic enzymes, transporters and cell wall degrading enzymes which are required for hyphal growth expansion during host colonization. Overall, these results suggest MoNdt80 mediated transcriptional regulation of GlcNAc catabolic pathway is essential for successful host colonization and pathogenesis.

Key message NCP1, a NINJA family protein lacking EAR motif, acts as a negative regulator of ABA signaling by interacting with and inhibiting the activity of transcriptional activator ABP9. The phytohormone abscisic acid plays a pivotal role in regulating plant responses to a variety of abiotic stresses including drought and salinity. Maize ABP9 is an ABRE-binding bZIP transcription activator that enhances plant tolerance to multiple stresses by positively regulating ABA signaling, but the molecular mechanism by which ABP9 is regulated in mediating ABA responses remains unknown. Here, we report the identification of an ABP9-interacting protein, named ABP Nine Complex Protein 1 (NCP1) and its functional characterization. NCP1 belongs to the recently identified NINJA family proteins, but lacks the conserved EAR motif, which is a hallmark of this class of transcriptional repressors. In vitro and in vivo assays confirmed that NCP1 physically interacts with ABP9 and that they are co-localized in the nucleus. In addition, NCP1 and ABP9 are similarly induced with similar patterns by ABA treatment and osmotic stress. Interestingly, NCP1 over-expressing Arabidopsis plants exhibited a reduced sensitivity to ABA and decreased drought tolerance. Transient assay in maize protoplasts showed that NCP1 inhibits the activity of ABP9 in activating ABRE-mediated reporter gene expression, a notion further supported by genetic analysis of drought and ABA responses in the transgenic plants over-expressing both ABP9 and NCP1. These data together suggest that NCP1 is a novel negative regulator of ABA signaling via interacting with and inhibiting the activity of ABP9.

Heterotrimeric G-protein a and beta-subunits regulate H2O2-mediated aerenchyma formation. The rice G-protein gamma-subunit, dense and erect panicle 1 (DEP1), is known to interact with the a-subunit and regulate nitrogen utilization and yield. However, it is unclear whether DEP1 regulates cell death for aerenchyma formation in rice roots. Using wild-type WYJ8 and its transgenic line WYJ8(DEPJ), we confirmed that DEP1 is involved in H2O2-mediated aerenchyma formation. The rates of aerenchyma formation varied in different parts of the roots in both varieties, with the highest rate in the 4-7 cm segments, reaching a plateau in the 7-8 cm segments. Compared with WYJ8, the aerenchyma area and H2O2 content in WYJ8(DEP1) were increased by 55.98% and 53.37%, respectively; however, the responses of aerenchyma formation to exogenous H2O2 were basically the same in the two varieties. Diphenylene iodonium (DPI) treatment had no effect on H2O2 production and elimination processes in WYJ8, but significantly reduced the activity of the key enzyme that catalyzes H2O2 biosynthesis in WYJ8(DEP1). Importantly, exogenous H2O2 treatment did not offset the effect of the decrease in endogenous H2O2 level caused by DPI on aerenchyma formation. These results indicated that DEP1 enhanced H2O2 biosynthesis and promoted the cell death of the root cortex, thus contributing to aerenchyma development in WYJ8(DEP1).

The insufficient intracellular H2O2 level in tumor cells is closely associated with the limited efficacy of chemodynamic therapy (CDT). Despite tremendous efforts, engineering CDT agents with a straightforward and secure H2O2 supplying ability remains a great challenge. Inspired by the balance of H2O2 generation and elimination in cancer cells, herein, a nanozyme-based H2O2 homeostasis disruptor is fabricated to elevate the intracellular H2O2 level through facilitating H2O2 production and restraining H2O2 elimination for enhanced CDT. In the formulation, the disruptor with superoxide dismutase-mimicking activity can convert O-2(center dot-) to H2O2, promoting the production of H2O2. Simultaneously, the suppression of catalase activity and depletion of glutathione by the disruptor weaken the transformation of H2O2 to H2O. Thus, the well-defined system could perturb the H2O2 balance and give rise to the accumulation of H2O2 in cancer cells. The raised H2O2 level would ultimately amplify the Fenton-like reaction-based CDT efficiency. Our work not only paves a way to engineer alternative CDT agents with a H2O2 supplying ability for intensive CDT but also provides new insights into the construction of bioinspired materials.

In this work, (Z)-N-benzoyl-N ‘-(1H-1,2,4-triazol-3-yl)carbamimidothioic acid and its Mn(II), Co(II), Cu(II) and Cd(II) complexes were introduced for the first time. This carbonyl thiourea ligand was prepared by the reaction of 1H-1,2,4-triazol-3-amine with benzoyl isothiocyanate. The structural elucidation of these compounds was performed using elemental analysis and spectral and magnetic measurements. Octahedral structures of all complexes, except Cd(II) complex with a tetrahedral geometry, were confirmed by applying DFT structural optimization. The thermal decomposition behaviour of metal complexes of carbonyl thiourea ligand is discussed. The calculation of kinetic parameters for prepared complexes (E-a, A, Delta H*, Delta S* and Delta G*) of all thermal degradation stages has been evaluated using two comparable approaches. Antimicrobial and ABTS-antioxidant studies indicated potent activity of Cd(II) complex compared with the other investigated compounds. The cytotoxic activity of the prepared compounds was investigated in vitro. The results indicated potent activity of Mn(II) complex against both HePG2 (liver carcinoma) and MCF-7 (breast carcinoma) cancer cells.

Bowman-Birk trypsin inhibitors (BBIs) play important roles in animal and plant immunity, but how these protease inhibitors are involved in the immune system remains unclear. Here, we show that the rice (Oryza sativa) BBI protein APIP4 is a common target of a fungal effector and an NLR receptor for innate immunity. The novel flame retarded unsaturated polyester resins have been developed and prepared by introduction of high nitrogen content additives into the polymer matrix in order to verify their effectiveness in the formation of swollen carbonaceous char inhibiting the burning process of the polymer. The intumescent flame retardants (IFRs) based on mixture or metal complex were developed and characterized by particle size distribution, Fourier-transform infrared spectroscopy (FTIR), nuclear magnetic resonance (NMR), powder X-ray diffraction (XRD), elemental analysis (CHN) and thermogravimetric analysis (TGA). The evaluation of the efficiency of IFRs addition on the flammability and smoke emission of the unsaturated polyester resins (UP) was carried out using the fire hazard (UL-94), limiting oxygen index (LOI) and cone calorimeter (CC) tests, as well as smoke density chamber tests. The volatile compounds evolved during the burning of materials were determined using a steady state tube furnace and a gas chromatograph with mass spectrometer. Furthermore, the prepared materials were subjected to differential scanning calorimetry (DSC), thermogravimetric analysis and water resistance tests. The mechanical properties of the materials were investigated using Shore D hardness and dynamic mechanical thermal analysis (DMA). The structural evaluation of the manufactured materials and samples after the cone calorimetry tests was carried out using scanning electron microscopy (SEM). It was found that the incorporation of new intumescent flame retardants led to the formation of carbonaceous char layers’ inhibiting the decomposition process and limiting the smoke emission. The most promising results were obtained for the resin containing complex designated as ZN3AT, for which the highest reduction in maximum values of heat release rate (419 kW/m(2)) compared to unmodified polymer (792 kW/m(2)) were recorded. Apart from that, the prepared intumescent flame retardants affect the cross-linking process as well as the thermal and mechanical properties of the UP.

Inorganic nanomaterials have gathered significant attention due to their biological enzyme-like catalytic activities, which are currently being utilized to display excellent biomedical applications. Among these, inorganic nanomaterials exhibiting catalase enzyme-like properties are of particular interest because they can impart protection to mammalian cells from cellular hydrogen peroxide (H2O2) and other free radicals. In this study, we have investigated the utility of bovine serum albumin (BSA) coated magnesium nanopartides (BSA-MgNPs) with respect to the protection of mammalian hepatic cells with depleted cellular catalase enzyme. We observed that BSA-MgNPs lead to the rapid breakdown of H2O2 and therefore protect mammalian cells exposed to 3-amino-1,2,4-triazole (3-AT), an irreversible catalase enzyme inhibitor. Results revealed that 3-AT induces oxidative stress by challenging the glutathione (GSH) and catalase enzyme, thus altering the cellular redox balance, which leads to the formation of reactive oxygen species (ROS) in human hepatocytes (WRL-68). Results showed that BSA-MgNPs impart protection to the catalase depleted hepatic cells. Expression analysis showed that BSA-MgNPs favorably modulate cellular antioxidant proteins/ enzymes and genes such as catalase, thioredoxin reductase (TrxR), peroxiredoxin 6 (Prx6), and Hsp 70 (heat shock protein). These results conclude that BSA-MgNPs impart protection to WRL-68 cells from acatalasia to display better cell survival as well as a remarkable decrease in the intracellular ROS levels.
