A convenient method for the synthesis of 2-(2-R-5-oxo-6,7-dihydro-4H-[1,2,4]triazolo-[1,5-a]pyrimidin-6-yl)acetanilides based on the regioselective domino reaction of N-arylitaconimides with substituted 3-aminotriazoles was proposed. Presumably, the reaction pathway includes the conjugated aza-addition of the endo-nucleophilic center of triazole to imide, followed by the recyclization of the intermediate to triazolo[1,5-a]pyrimidine.

Based on the previous studies that revealed the valuable role of pyrazole scaffold in cancer management and VEGFR-2 inhibition, a new set of pyrazole conjugated with pyrazoline, triazolopyrimidine and pyrazolone moieties were synthesized and investigated for their anticancer efficiency against human breast cancer MCF-7. The anticancer screening revealed the significant sensitivity of breast carcinoma towards compounds 4b, 5c, 6c, 7b, 7c and 12c with IC50 values ranging from 16.50 – 26.73 mu M in comparison with tamoxifen (IC50 = 23.31 mu M). Moreover, the new analogues were further examined for their VEGFR-2 inhibitory activity, among the tested derivatives 5c, 6c, 7b, 7c and 12c displayed prominent inhibitory efficiency versus VEGFR-2 kinase with % inhibition ranging from 70 to 79%. Compounds 6c, 7c and 12c revealed inhibitory efficiency in nanomolar level with IC50 (913.51, 225.17 and 828.23 nM, respectively) comparing to sorafenib (IC50 = 186.54 nM). Flow cytometric analysis revealed that the promising compound 12c prompted pre-G1 apoptosis and cell growth cessation at G2/M phase and stimulated apoptosis via activation of caspase-3. Moreover, molecular docking study of the promising derivatives was performed to highlight their binding modes and interactions with the amino acid residues of VEGFR-2 enzyme.

Oxidative stress biomarkers are powerful endpoints in toxicological research. Cellular reductive/oxidative balance affects numerous signaling pathways involving H2O2. Detoxification and control of H2O2 levels results mainly from catalase activity. The aim of this work was to develop a precise, simple, cost-effective microassay to measure catalase activity in small tissue samples and cell extracts. We developed a protocol that quantifies H2O2 decomposition by intrinsic catalase in biological samples. Catalase activity was calculated based on rate of decomposition of H2O2, following absorbance at 240 nm. We developed a multi-well spectroscopic approach, reducing sample quantity requirements and allowing simultaneous assessment of large number of samples. The protocol is sensitive across a wide range of catalase activity (11.5-7575 U). The assay presents a 95% confidence interval with an intra-assay coefficient of variation of 3.7%, an inter-assay coefficient of variation of 6.2% and good correlation with a commercial kit. The assay was established and validated for different biological samples, including sheep hepatic tissue and human tumor and non-tumor cell lines. This high-throughput method is robust, sensitive, time-saving and cost-effective, generating highly reproducible results with precision and good correlation with a commercial kit reinforcing the method’s validity for research and toxicological applications.

Herein, Fe3O4 nano-particles are anchored over nitrogen doped reduced graphene oxide Fe3O4@N-rGO) structure to accommodate electric and magnetic components, so that it could match both the conduction and polarization loss to augment the attenuation of electromagnetic radiation. The formation of condensed heteroaromatic structure and decoration of Fe3O4 particles on r-GO structure are confirmed by photoelectron spectroscopy (XPS). high-resolution transmission electron microscopy (HRTEM), X-ray diffraction (XRD). The Fe3O4@N-rGO hybrid is subsequently dispersed in epoxy matrix, which result in significantly higher electrical conductivity (1.31 s/m) and electromagnetic interference (EMI) shielding efficiency (- 26 dB at 1 mm thickness). It was assumed that presence of electric-magnetic integrated Fe3O4@N-rGO hybrid in epoxy matrix significantly improved the interfacial polarization and anisotropy energy, which was reflected in absorption loss value of -27 dB. The composites also exhibited enhanced mechanical and thermo-mechanical properties coupled with improved thermal stability.

In our approach to synthesize bioactive molecules, a series of novel N-heterocycles were synthesized and evaluated for their in vitro antitumor activity against a panel of three human cancer cell lines, namely, human breast cancer cell line (MCF-7), human cervical cancer cell line (HeLa), and human prostate cancer PC-3. The majority of the tested compounds exhibited significant cytotoxic activity toward the tested tumor cell lines. Analogues 33, 34, 31, 38, 21, 23, 22, and 20 exhibited considerable cytotoxic activities comparable with standard drug 5-fuorouracil. Compound 33 displayed superior cytotoxicity with IC50 value of 4.12 +/- 1.21 mu g/mL against HeLa tumor cell line.

A series of new substituted benzimidazoles embedded with a variety of function groups has been synthesized from N-methyl-2-bromoacetylbenzimidazole. The synthesized compounds were fully characterized, and their structures were elucidated based on elemental analysis, spectral data, and alternative synthetic pathways, whenever possible. In this work, seven small molecular compounds named N,N-diphenyl-4-(2-phenylpyrimido [1,2-b]indazol-4-yl) aniline (1), N,N-diphenyl-4-(5-phenylpyrazolo [1,5-a]pyrimidin-7-yl)aniline (2), N,N-diphenyl-4-(5-phenyl[1,2,4]triazolo [1,5-a]pyrimidin-7-yl)aniline (3), 4,4′-(pyrimido [1,2-b]indazole-2,4-diyl)bis (N,N-diphenylaniline) (4), 4,4′-(pyrazolo [1,5-a]pyrimidine-5,7-diyl)bis (N,N-diphenylaniline) (5), 4,4′-([1,2,4]triazolo [1,5-a] pyrimidine-5,7-diyl)bis (N,N-diphenylaniline) (6), N,N-diphenyl-4-(4-phenylpyrimido [1,2-b]indazol-2-yl)aniline (7) with donor-acceptor (D-A) structure were designed and synthesized by employing triphenylamine (TPA) as donor and indazole, pyrazole and/or triazole as acceptor. Systematic study and analysis on thermal, photo physical, electrochemical properties of all compounds were performed. All compounds exhibit good thermal and morphological stabilities with decomposition temperatures (T-d) range from 270 to 462 degrees C, and glass transition temperatures (T-g) range from 78 to 127 degrees C. The investigation of photophysical properties of these compounds revealed that all these compounds are of high photoluminescence quantum yields (PLQY), particularly for compound 6, whose PLQY value was as high as 90.22%. To explore potential applications of these materials in organic light-emitting diodes (OLEDs), two devices with compounds 2 and 5 were fabricated. Both devices show excellent device performances with maximum current efficiency, maximum power efficiency and external quantum efficiency of 15.9 cd A(-1), 10.41 m W-1 and 8.4% for compound 2-based device and 10.1 cd A(-1), 5.81 m W-1 and 5.5% for compound 5-based device, respectively.

Ultrathin C3N5 nanosheets with enhanced photocatalytic methylene blue (MB) degradation and H-2-evolution performance were prepared from thermal treatment of 3-amino-1,2,4-triazole (3-AT) and NH4Cl followed with a protonate procedure. The characterization results revealed that the protonating process could contribute to the exfoliation of C3N5 with large surface area, the effective charge transfer capability and the modified band structure. The as-prepared C3N5 nanosheets exhibited enhanced properties in photocatalytic reactions such as MB photodegradation and H-2-evolution from water splitting. This study offered a feasible route to prepare highly-efficient two-dimensional photocatalyst, which could be applied potentially for implementation in wide range of energy generation and environmental applications.

Development of the neurovascular unit (NVU) is a complex, multistage process that requires orchestrated cell signaling mechanisms across several cell types and ultimately results in formation of the blood-brain barrier. Typical high-throughput screening (HTS) assays investigate single biochemical or single cell responses following chemical insult. As the NVU comprises multiple cell types interacting at various stages of development, a methodology combining high-throughput results across pertinent cell-based assays is needed to investigate potential chemical-induced disruption to the development of this complex cell system. To this end, we implemented a novel method for screening putative NVU disruptors across diverse assay platforms to predict chemical perturbation of the developing NVU. HTS assay results measuring chemical-induced perturbations to cellular key events across angiogenic and neurogenic outcomes in vitro were combined to create a cell-based prioritization of NVU hazard. Chemicals were grouped according to similar modes of action to train a logistic regression literature model on a training set of 38 chemicals. This model utilizes the chemical-specific pairwise mutual information score for PubMed MeSH annotations to represent a quantitative measure of previously published results. Taken together, this study presents a methodology to investigate NVU developmental hazard using cell-based HTS assays and literature evidence to prioritize screening of putative NVU disruptors towards a knowledge-driven characterization of neurovascular developmental toxicity. The results from these screening efforts demonstrate that chemicals representing a range of putative vascular disrupting compound (pVDC) scores can also produce effects on neurogenic outcomes and characterizes possible modes of action for disrupting the developing NVU.

Nowadays, the research of proton conductive materials has been extended from traditional sulfonated polymers to novel crystalline solid materials. It is important but still challenging to develop high-performance proton-conducting materials suitable for proton exchange membrane fuel cells (PEMFCs). Considering the features of molybdenum oxide and triazole in the construction of proton-conducting material, two triazole-modified molybdenum oxides, [MoO3(trz)(0.5)]1and [H(atrz)(4)[(atrz)(2)(gamma-Mo8O26)] . 2H(2)O2(trz=1,2,4-triazole, atrz=3-amino-1,2,4-triazole), were re-synthesized under optimized synthetic conditions, and their proton-conducting performances were investigated.1and2show high proton conductivities of 1.94×10(-2) S cm(-1)(85 degrees C, 95 % RH) and 1.3×10(-3) S cm(-1)(65 degrees C, 95 % RH), respectively.
