One of the main directions of steroid research is the preparation of modified derivatives in which, in addition to changes in physicochemical properties, receptor binding is significantly altered, thus a bioactivity different from that of the parent compound predominates. In the frame of this work, 2-arylidene derivatives were first synthesized by regioselective modification of the A-ring of natural sex hormone, 5 alpha-dihydrotestosterone (DHT). After Claisen-Schmidt condensations of DHT with (hetero)aromatic aldehydes in alkaline EtOH, heterocyclizations of the alpha,beta-enones were performed with 3-amino-1,2,4-triazole, 3-aminopyrazole and 3-amino-5-methylpyrazole in the presence of t-BuOK in DMF to afford 7 ‘-epimeric mixtures of A-ring-fused azolo-dihydropyrimidines, respectively. Depending on the electronic demand of the substituents of the arylidene moiety, spontaneous or 2,3-dichloro-5,6-dicyanobenzoquinone (DDQ)-induced oxidation of the heteroring led to triazolo[1,5-a]pyrimidines and pyrazolo[1,5-a]pyrimidines in good yields, while, using the Jones reagent as a strong oxidant, 17oxidation also occurred. The crystal structures of an arylidene and a triazolopyrimidine product have been determined by single crystal X-ray diffraction and both were found to crystallize in the monoclinic crystal system at P21 space group. Most derivatives were found to diminish the transcriptional activity of androgen receptor (AR) in reporter cell line. The candidate compound (17 beta-hydroxy-2-(4-chloro)benzylidene-5 alpha-androstan-3-one, 2f) showed to suppress androgen-mediated AR transactivation in a dose-dependent manner. We confirmed the cellular interaction of 2f with AR, described the binding in AR-binding cavity by the flexible docking and showed the ability of the compound to suppress the expression of AR-regulated genes in two prostate cancer cell lines.

Herein, a novel and sensitive fluorescence method for cartap determination is established on the basis of the inner filter effect (IFE) of gold nanoparticles (Au NPs) on the fluorescence of glutathione protected Cu NCs (GSH-Cu NCs). In the presence of Au NPs, the fluorescence of GSH-Cu NCs was strongly quenched by the IFE because the absorption spectra of Au NPs overlap well with the emission spectra of GSH-Cu NCs. Upon addition of cartap, cartap could induce the aggregation of Au NPs whose absorption spectrum does not overlap with the emission spectrum of GSH-Cu NCs. Then, with the increase in cartap concentration, the IFE-decreased fluorescence was gradually recovered, realizing the fluorescence sensing of cartap. Under optimal conditions, the proposed method has a good linear relationship with cartap concentration in the range of 7-100 nM, and the detection limit is 3.34 nM. In addition, satisfactory results were obtained for cartap analysis using tap water and cabbage as real samples, which demonstrated that the method as-developed would have great practical application prospects.

Herein, an effectual, quick and novel method is described for the synthesis of new triazolo[1,5-a]pyrimidine, triazolo[5,1-b][1,3] thiazine and pyrazolo[1,5-a]pyrimidine derivatives. This series of fused six-member rings to triazole and pyrazole was prepared via the catalyst-free reaction of dialkyl acetylenedicarboxylates and 3-substituted 1H-1,2,4-triazole or 3-amino-1H-pyrazole-4-carbonitrile. The structures of the prepared products were deduced from their Fourier-transform infrared, elemental analysis and proton and carbon-13 nuclear magnetic resonance spectral data. Wood (secondary xylem) formation in tree species is dependent on auxin-mediated vascular cambium activity in stems. However, the complex regulatory networks underlying xylem formation remain elusive. Xylem development in Populus was characterized based on microscopic observations of stem sections in transgenic plants. Transcriptomic, quantitative real-time PCR, chromatin immunoprecipitation PCR, and electrophoretic mobility shift assay analyses were conducted to identify target genes involved in xylem development. Yeast two-hybrid, pull-down, bimolecular fluorescence complementation, and co-immunoprecipitation assays were used to validate protein-protein interactions. PaC3H17 and its target PaMYB199 were found to be predominantly expressed in the vascular cambium and developing secondary xylem in Populus stems and play opposite roles in controlling cambial cell proliferation and secondary cell wall thickening through an overlapping pathway. Further, PaC3H17 interacts with PaMYB199 to form a complex, attenuating PaMYB199-driven suppression of its xylem targets. Exogenous auxin application enhances the dual control of the PaC3H17-PaMYB199 module during cambium division, thereby promoting secondary cell wall deposition. Dual regulation of xylem formation by an auxin-mediated PaC3H17-PaMYB199 module represents a novel regulatory mechanism in Populus, increasing our understanding of the regulatory networks involved in wood formation.

This work introduces a new additive named 4,4′-trimethylenedipiperidine for the practical and ecofriendly preparation of ethyl 5-amino-7-(4-phenyl)-4,7-dihydro-[1,2,4]triazolo[1,5-a]pyrimidine-6-carboxylate derivatives. This chemical is commercially available and easy to handle. It also possesses a low melting point and a broad liquid range temperature, high thermal stability, and good solubility in water. Based on green chemistry principles, the reaction was performed in a) a mixture of green solvents i.e. water and ethanol (1:1 v/v) at reflux temperature, and b) the additive was liquefied at 65 degrees C and the reaction was conducted in the liquid state of the additive. High yields of the desired triazolo-pyrimidines were obtained under both aforementioned conditions. Our results demonstrated that this additive, containing 2 Lewis base sites and able to act as an acceptor-donor hydrogen bonding group, is a novel and efficient alternative to piperidine, owing to its unique properties such as its reduced toxicity, nonflammable nature, nonvolatile state, broad liquid range temperature, high thermal stability, and ability to be safely handled. Furthermore, this additive could be completely recovered and exhibited high recyclability without any change in its chemical structure and no significant reduction in its activity. The current methodology has several advantages: (a) it avoids the use of hazardous materials, as well as toxic, volatile, and flammable solvents, (b) it does not entail tedious processes, harsh conditions, and the multistep preparation of catalysts, (c) it uses a metal-free and noncorrosive catalyst, and (d) reduces the generation of hazardous waste and simple work-up processes. The most important result of this study is that 4,4′-trimethylenedipiperidine can be a promising alternative for toxic, volatile, and flammable base reagents in organic synthesis owing to its unique properties.

In this work, amine-functionalized ZIF8 (i.e., ZIF8-A) was dispersed in n-heptanol (ZIF8-A/n-heptanol) to remove SO2. The effects of ZIF8-A proportion, SO2 concentration, and absorption and desorption temperatures on the removal of SO2 were systematically investigated. The experimental results showed that the activity of ZIF8-A on SO2 removal was enhanced after being dispersed in n-heptanol. The SO2 saturation capacity increased with increasing proportion of ZIF8-A and SO(2)concentration. The optimal temperatures for SO2 absorption and desorption were found to be 25 and 120 degrees C, respectively. Under the optimal experimental conditions (25 degrees C and 4% SO2), the SO2 saturation capacity was 589 mg/g. Five continuous absorption-desorption cycles suggested a good regeneration performance. Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, ultraviolet analyses, and computational simulation indicated that SO2 mainly interacted with ZIF8-A via hydrogen bonds besides physical absorption. The simulation results further indicated that the enhanced SO2 adsorption performance of ZIF8-A when dispersed in n-heptanol was due to the lower self-diffusion coefficient and higher isosteric heat of SO2 molecules in ZIF8-A/nheptanol.

A series of 1,2,4-triazolo [1,5-a]pyrimidine derivatives was designed, synthesized and screened for their antibacterial and antifungal activities as well as their safety profile. Compounds 2b, 3a, 6b, 8b, 8c, 8h, 9a,b, 10b, 11a,b and 12a,b showed high activity against Gram-positive and Gram-negative bacteria with MIC values ranging from 0.25 to 2.0 mu g/mL. Many compounds were safe with no cytotoxicity against human embryonic kidney and red blood cells at concentration up to 32 mu g/mL. Moreover, compound 9a showed the highest inhibitory activity against DNA Gyrase with IC50 = 0.68 mu M compared to ciprofloxacin IC50 = 0.85 mu M. Molecular docking at DNA Gyrase active site revealed binding mode and docking scores comparable to that of ciprofloxacin confirming their antibacterial activity via DNA Gyrase inhibition.

Development of the neurovascular unit (NVU) is a complex, multistage process that requires orchestrated cell signaling mechanisms across several cell types and ultimately results in formation of the blood-brain barrier. Typical high-throughput screening (HTS) assays investigate single biochemical or single cell responses following chemical insult. As the NVU comprises multiple cell types interacting at various stages of development, a methodology combining high-throughput results across pertinent cell-based assays is needed to investigate potential chemical-induced disruption to the development of this complex cell system. To this end, we implemented a novel method for screening putative NVU disruptors across diverse assay platforms to predict chemical perturbation of the developing NVU. HTS assay results measuring chemical-induced perturbations to cellular key events across angiogenic and neurogenic outcomes in vitro were combined to create a cell-based prioritization of NVU hazard. Chemicals were grouped according to similar modes of action to train a logistic regression literature model on a training set of 38 chemicals. This model utilizes the chemical-specific pairwise mutual information score for PubMed MeSH annotations to represent a quantitative measure of previously published results. Taken together, this study presents a methodology to investigate NVU developmental hazard using cell-based HTS assays and literature evidence to prioritize screening of putative NVU disruptors towards a knowledge-driven characterization of neurovascular developmental toxicity. The results from these screening efforts demonstrate that chemicals representing a range of putative vascular disrupting compound (pVDC) scores can also produce effects on neurogenic outcomes and characterizes possible modes of action for disrupting the developing NVU.

Nowadays, the research of proton conductive materials has been extended from traditional sulfonated polymers to novel crystalline solid materials. It is important but still challenging to develop high-performance proton-conducting materials suitable for proton exchange membrane fuel cells (PEMFCs). Considering the features of molybdenum oxide and triazole in the construction of proton-conducting material, two triazole-modified molybdenum oxides, [MoO3(trz)(0.5)]1and [H(atrz)(4)[(atrz)(2)(gamma-Mo8O26)] . 2H(2)O2(trz=1,2,4-triazole, atrz=3-amino-1,2,4-triazole), were re-synthesized under optimized synthetic conditions, and their proton-conducting performances were investigated.1and2show high proton conductivities of 1.94×10(-2) S cm(-1)(85 degrees C, 95 % RH) and 1.3×10(-3) S cm(-1)(65 degrees C, 95 % RH), respectively.
