A series of fluconazole analogs containing a 1,2,3-triazole fragment has been designed and synthesized on the basis of the active site of the cytochrome P 450 14α-demethylase (CYP51). Their structures were characterized by 1H NMR, 13C NMR, and LC-MS. The MIC80 values indicate that the target compounds showed higher activities against nearly all the fungi tested to some extent except against Aspergillus fumigatus. Several compounds, e.g. I, showed 128 times higher activity (with the MIC80 value of 0.0039 mg/mL) than that of fluconazole against Candida albicans and also showed higher activity than that of the other pos. controls.

To study the antifungal mechanism and structure-activity relationship of the azole compounds, four triazole compounds were designed and synthesized. The said triazole derivatives, e.g., I, were prepared via Friedel-Crafts acetylation of 1,3-difluorobenzene with chloroacetyl chloride; the resulting α-chloro-2,4-difluoroacetophenone underwent condensation with 1,2,4-triazole to give α-(1,2,4-triazol-1-yl)-2,4-difluoroacetophenone, which underwent cyclization to give 1-[2-(2,4-difluorophenyl)-1,2-epoxy-3-propyl]-1H-1,2,4-triazole, which underwent additive ring-opening with Et 1-piperazinecarboxylate to give 1-(1H-1,2,4-triazol-1-yl)-2-(2,4-difluorophenyl)-3-(4-ethoxycarbonylpiperazin-1-yl)propan-2-ol, which underwent hydrolysis to give 1-(1H-1,2,4-triazol-1-yl)-2-(2,4-difluorophenyl)-3-(piperazin-1-yl)propan-2-ol, which underwent N-alkylation with alkyl halides to give the title compounds All of them were confirmed by 1H-NMR and MS spectra. Antifungal activities of title compounds were evaluated. And the results of the preliminary antifungal test showed that all compounds exhibited inhibitory effects on C. albicans and C. neoformans. Two of the compounds showed higher activity than fluconazole.

To study the effect of triazole derivatives with a side chain containing a 4-substituted-1-piperidinyl groups or a similar secondary amine on the antifungal activity of triazole compounds, nine title compounds were synthesized and determined by elementary anal., 1H-NMR and IR spectra. An example compound thus prepared was α-(2,4-difluorophenyl)-α-(1H-1,2,4-triazol-1-ylmethyl)-1-piperidineethanol. The MICs of all the title compounds were determined by a method recommended by the national committee for clin. laboratory standards (NCCLS) using the RPMI 1640 test medium. All of the title compounds displayed potent antifungal activities to some extent. The activity of the two compounds were more than 4 times as high as that of fluconazole and itraconazole against Candida albicans in vitro. The stereochem. had important influence on the antifungal activities of the title compounds (no data).

In an attempt to find novel azole antifungal agents with improved activity and broader spectrum, computer modeling was used to design a series of new azoles with piperidin-4-one O-substituted oxime side chains. Mol. docking studies revealed that they formed hydrophobic and hydrogen-bonding interactions with lanosterol 14α-demethylase of Candida albicans (CACYP51). In vitro antifungal assay indicates that most of the synthesized compounds showed good activity against tested fungal pathogens. In comparison with fluconazole, itraconazole and voriconazole, several compounds (such as I, R = H, 3-Cl, 2-F) show more potent antifungal activity and broader spectrum, suggesting that they are promising leads for the development of novel antifungal agents.

Novel potential antifungal compound of 1,2,4-triazole class – 3,3′-(piperazine-1,4-diyl)bis(2-(2,4-difluorophenyl)-1-(1H-1,2,4-triazol-1-yl)propan-2-ol) (S-119) – has been synthesized and characterized. Solubility in buffer solutions (pH 2.0 and 7.4), 1-octanol, n-hexane, and ethanol was measured by the isothermal saturation method in the temperature range (293.15-313.15 K). The following order of the solubility was revealed: ethanol > buffer pH 2.0 > 1-octanol > buffer pH 7.4 > n-hexane. The solubility was correlated by the Van’t Hoff and Apelblat equations. The Hansen solubility parameters were applied to solubility evaluation and demonstrated the consistency with the exptl. data. The thermodn. parameters of solubility and transfer processes were calculated and discussed in view of solute-solvent interactions. The temperature dependences of the distribution coefficients of S-119 in 1-octanol/buffer and n-hexane/buffer (pH 2.0 and 7.4) two-phase systems were obtained and considered from the thermodn. point. The ΔlogD parameter determined from the distribution experiment clearly demonstrated the preference of lipophilic delivery pathways for S-119 in the biol. media and unfavorable transition to the non-polar regions. Membrane permeability coefficient of S-119 was determined and evaluated with the assistance of the resp. parameter for the structurally related drug fluconazole.
