The crude product was purified by column chromatography on silica gel 100-200 mesh using 50% EtOAchexane to afford the title compounds 3 and 3a. LCMS (mlz): 205.00 (M + 1).

The combined organic layers were dried, and concentrated The residue was purified by FC on S1O2 column (eluent : from DCM to DCM:MeOH 90:10) to afford 2-fluoro-6-(2H-1 ,2,3-triazol-2-yl)benzoic acid (p186, 1.1 g, y= 56%). Once the aldehyde was added the reaction was allowed to warm to room temperature. After 3 hours, the reaction was quenched by pouring it into a saturated solution of aqueous NH4C1. The aqueous portion was extracted with EtOAc in portions, 7 x 400 mL, The combined organics were washed with brine, dried over MgS04, filtered and concentrated to afford a brown oil. Chromatography on silica gel (10%) acetone-DCM increasing to 50% acetone and increasing to 10% MeOH-DCM) provided the title compound as an amber solid.

To a mixture of the product from step 2 (7.9 g, 38 mmol), cesium carbonate (24.8 g,76 mmol) and Cul (2.88 g, 7.6 mmol) in DMF (200 mL) were added 2H-[1,2,3]triazole (5.24 g,76 mmol) and N,N’-dimethyl-cyclohexane-l12-diamine (0.9 g, 6.5 mmol) and the mixture was heated to 110C overnight. The cooled reaction mixture was adjusted to ~pH 12 with 1M sodium hydroxide and extracted with EtOAc (50 mL x 3). The aqueous layer was adjusted to ~pH 4 with 1M HCl and extracted with EtOAc (50 mLx4). The extracts was dried over Na2SO4, filtered, the filtrate concentrated in vacuo and the residue purified by chromatography on silica (petroleum ether : EtOAc =10:1) to provide the title compound. LRMS m/z (M+H) 196.0 found, 196.0 required.

The reaction mixture was stirred at 85 oC for 16 h in a sealed tube. After consumption of the starting material (monitored by TLC), the reaction mixture was diluted with water (100 mL) and extracted with EtOAc (2 x 50 mL). The combined organic extracts were dried over sodium sulfate, filtered and concentrated in vacuo. The crude material was purified by column chromatography using 15-25% EtOAc:hexane to afford 1-fluoro-2-methoxy-4-nitrobenzene (1.2 g, 35%) as an off-white solid. 1H-NMR (CDCl3, 400 MHz): delta 8.87 (s, 1H), 8.10 (d, 1H), 8.00-7.97 (m, 2H), 4.10 (s, 3H), 2.50 (s, 3H); LCMS: 234.9 (M+1); (column; Ascentis Express C-18 (50 ¡Á 3.0 mm, 2.7 mum); RT 2.02 min. 0.025% Aq TFA+5% ACN: ACN+5% 0.025% Aq TFA; 1.2 mL/min); TLC: 30% EtOAc:hexane (Rf: 0.2).

The reaction mixture is stirred at 100 C for 2 hours. The reaction mixture is allowed to cool to room temperature, diluted with CH2C12 (100 mL), and washed with H20 (75 mL x2). The organic layer is dried with Na2S04 and filtered. The filtrate is concentrated under reduced pressure and the residue is purified by silica gel flash column chromatography with 5% MeOH in EtOAc as the eluent to afford an impure mixture of 5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-benzaldehyde plus regiosiomer (270 mg).To a mixture of 4-(2-amino-pyrimidin-5-yl)-Nl-ieri-butyl-benzene-l,2-diamine (270 mg, 1.05 mmol) and impure mixture of 5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-benzaldehyde plus regiosiomer (270 mg) in DMF (15.0 mL) is added Oxone (650 mg, 1.06 mmol) in H20 (5.0 mL). The reaction mixture is stirred at room temperature for 1.5 hours and then quenched with saturated sodium thiosulfate aq. (25 mL). The quenched reaction mixture is extracted with EtOAc (25 mL x2) and the combined organic layers are washed with H20 (50 mL x2), dried with Na2S04 and filtered. The filtrate is concentrated under reduced pressure and the residue is purified by silica gel flash column chromatography with 5% MeOH in CH2C12 as the eluent to afford an impure mixture of 5-(2-amino- pyrimidin-5-yl)- l-ieri-butyl-2-[5-chloro-2-(3-methyl- l,2,4-triazol-l-yl)-phenyl]-2,3- dihydro-lH-benzimidazol-2-ol plus regiosiomer (25 mg).

The mixture was stirred at ambient temperature over night. Hydrochloric acid (cone, 2.0 mL, 66 mmol) was added. The solid was isolated by filtration, washed with water and dried in vacuum to yield the title compound as a solid (8.3 g, 65%). MS (ESI ) m/z 224 [M-H]”.
